Q

What is the first-pass effect?

16
Q

What occurs during pharmacokinetic distribution?

A

-transport of drug from bloodstream to site
-unbound drugs are free to distribute and reach site

17
Q

What happens to drugs bound to plasma proteins?

A

they are pharmacologically inactive

18
Q

When does elimination start?

A

when drugs enter the bloodstream (primarily liver and kidneys)

19
Q

What organ is mostly responsible for pharmacokinetic metabolism?

20
Q

What does pharmacokinetic metabolism convert drugs into?

A

-water-soluble molecules (preparation for excretion)
-inactive metabolite
-less active metabolite
-more potent metabolite (prodrug)

21
Q

What organ is primarily responsible for excretion?

22
Q

What are some drugs that do not go through the first pass effect?

A

-orally disintegrating tablets (ODT)
-oral soluble films
-sublingual
-buccal
-some rectal

23
Q

Why do some oral drugs not go through first pass effect?

A

the oral cavity is highly vascularized

24
Q

What is the onset of action?

A

time for the drug to start physiologic response

25
What is peak effect?
time required for drug to meet max. therapeutic response
26
What is duration of action?
length of time drug concentration is sufficient to illicit therapeutic response
27
What is half-life?
time required for 50% of drug to be removed by the body
28
What is pharmacodynamics?
drug-induced physiological changes
29
What is the pharmacodynamic mechanism of action?
effect based on characteristics of cells/tissues targeted by the drug
30
What is the difference between an agonist and an antagonist?
-agonist= bind to receptors and activate them -antagonist= bind to receptors but do not activate them
31
What is pharmacotherapy?
use of drugs to prevent or treat diseases
32
How does intake of dark leafy greens impact drug effects?
may decrease anticoagulant effect (warfarin)
33
How does intake of grapefruit impact drug effects?
may cause problems with enzymes and transporters --> too much or too little of a drug (cardiac; antiseizure; anticholesterol; antianxiety med)
34
How does valerian root impact drug effects?
can increase drowsiness and sedation with CNS depressants
