Motility indole ornithine (Gram -)
Semi-solid )tempered medium
Motility?
Turbidity away from stab line
Does bacteria have tryptophanase?
If present, indole pyruvic acid formed from broken down tryptophan
Indole deamination
When pH moved to acidic, indole pyruvic acid separated into indole + pyruvic acid
Kovac’s reagent
Reacts with indole, forms red ring at top of medium
Ornithine
Does bacteria possess ornithine decarboxylase?
Decarboxylation loses CO2, putrescine
Putrescine produced = raised pH = turns purple when pH > 7.6
Bromocresol purple
Glucose turns media yellow, then back to purple. If stays yellow, glucose metabolized and putrescine not produced.
Ingredients
Tryptophan
Kovac’s reagent
