Triple sugar iron (Gram -)
Can it utilize sugars: glucose (0.1%), lactose (1.0%), sucrose (1.0%)
Ingredients
Glucose
Lactose
Sucrose
Phenol red
Peptone
Thiosulfate
Ferric ammonium citrate
Phenol red (lactose metabolized?)
pH < 6.8
Peptone (partially hydrolyzed)
If peptone used, ammonia produced = pH back to alkaline = back to red
CO2 released (decarboxylation)
Gas pocket present, krebs cycle
Thiosulfate
Sugar metabolized = acidic environment = sulfur released
If sulfur can act as terminal electron acceptor
Product is H2S (hydrogen sulfide)
Ferric ammonium citrate
Binds with H2S = black color
