how do proteins interact with ligands
they form an ezyme substrate complex in an equilibrium reaction
what is ka?
rate of association
what is kb?
rate of dissociation
what is Ka?
ka/kd: The association constant, it tells you the affinity your enzyme has towards its ligan
what does the association constant tell you about the equilibrium of an enzyme and its ligand?
a Ka>1 tells you that association is favored and there is strong affinity between the enzyme and its ligand
if Ka<1, if tells you that dissociation is favored and there is poor affinity between the enzyme and its ligand
what is the occupancy rate?
Y= the total binding sites occupied (PL)/ total protein (PL+P)
What is Kd?
it is 1/Ka
what does it mean if Kd is small?
it means that Ka is high, meaning that the enzyme has strong affinity for its subrate and disassociation is slow
what does it mean if kd is big?
it means that Ka is low, meaning that the enzyme has low affinity for its substrate and the dissociation rate is fast
what does it mean if the occupancy rate is 0.5?
it means that 1/2 of the total binding sites are occupied
