21
Q

What is strepto-

22
Q

What is gram variability?

A

When some cells of a pure culture appear red and some appear purple.

23
Q

When is gram variability most likely to occur?

A

In older cultures

24
Q

What color are Gram- positive organisms?

25
What color are Gram- negative organisms?
Red/pink
26
Why are Gram- positives purple?
They have a thick Peptidoglycan layer and do not have an outer envelope so the Crystal violet stays in the cell
27
Why are gram negative red?
They do have an outer envelope that dissolve when the alcohol is used. The alcohol removes the Crystal violet so they stain red with Safranin Counterstain.
28
Which genus organism is categorized as acid-fast?
Mycobacterium
29
Does acid bath require heat in the form of steam bath?
Yes
30
What color are acid fast organisms?
Red/pink
31
What color are non-acid fast organisms
Blue
32
Does spore staining require heat in the form of a steam bath?
Yes
33
What is the color of the bacterial cell in spore staining?
Red/pink
34
What color is the spore in spore staining?
Green
35
What is the function of a bacterial spore?
For when their are unfavorable conditions for the bacteria
36
Is the slide heat fixed when doing capsule staining?
No
37
What is the color of the bacterial cell in capsule staining?
Purple
38
What is the color of the capsule in capsule staining?
Clear halo
39
What is the function of a capsule?
The capsule protects the cell against host immune system.
40
What are the ingredients for spore staining?
1. Brilliant green 2. Di water 3. Safranin
41
Steps for spore staining
1. Flood with green over heat for 3 mins 2. Cool for 5 mins 3. Counterstain with Safranin 30 secs 4. Di water 5. Blot
