The basis of Voges-Proskauer testing is that acetyl-methyl carbinol is produced from glucose and it is then converted to diacetyl in the presence of alpha naphthol and atmospheric oxygen. The diacetyl and the peptones found in the broth condense to form a pink/red polymer.
